Now I copied to hard drive as Disc Image File but file is 4.38 GB size.
How can I make it smaller file from CloneDVD2?

Missed Slysoft and thank you for all the work enjoyed more 10years(?).

Near the bottom of CloneDVD2's Title Configuration page is a section to select Quality

Use the drop down arrow to choose DVD-5

That should reduce the size down to 4.36 GB so that it will fit on a single layer dvd
 
Sorry, what I mean make file smaller such as mp4 or get it 500k.

You don't with CloneDVD, portable files such as mp4 are created by 'CloneDVD Mobile'. Oh and just an fyi, if you created the dvd iso with the anydvd ripper, please don't.
